Brief Information About Collagenase

Collagenases are enzymes calledendopeptidases that break the peptide bonds or triple helix region in native collagen. Collagen is an important component of extracellular connective tissue matrix of the animals which is made by breaking of pro-collagen by collagenase after it is secreted by the cell.  This process prevents the large structures from being formed inside the cell. Apart from being produced by bacteria, normal immune response of the body also produces it.

The difference between bacterial collagenases and vertebrate collagenases is that bacterial collagenases show broader substrate specificity and unlike animal collagenases which splitthe triple-helical conformation of collagen,bacterial collagenases degrade both water-soluble denatured collagens and water-insoluble native ones. In addition to being produced by some bacteria, collagenase can be made by the body as part of its normal immune response.

In 1950s the pioneering work by Mandl, Harper, Seifter and their fellow associates, and classification work of Van Wart and Bond led to the discovery of characteristics of clostridial collagenase. In the year 1959, collagenase was first made commercially available by isolating it from Clostridium histolyticum. The work was done by Worthington. In 1962, Gross and Lapiere got proof of collagenase in culture media of bullfrog tadpole tissue which led to discovery of first vertebrate collagenase. After this many collagenases were found in mammals, amphibians, marine life and bacteria. Till then all research on collagenases were based on clostridial collagenases. The most significant of researches after that was the discovery of between collagenase and diseases like metastasis, wound healing, tissue repair, angiogenesis, herniated disc treatment, rheumatoid arthritis etc.

Latest applications of collagenase:

  • Isolation of hepatocyes, lung cells and adipocytes, from adrenal tissue, lung and epithelium.
  • Isolation of cardiomyocytes from muscle, cartilage, bone, thyroid, and endothelium
  • Isolation of and various other soft tissues
  • Isolation of mammary tissue,porcine and human pancreatic islet cells
  • Treatment of Dupuytren's contracture andXiaflex or Peyronie's disease
